Summary

Pyrrolidine-3-Carboxylic Acid Derivatives (Beta-Proline Derivatives) having potential therapeutic applications.

The global proline market size is currently about USD 268 million and is projected to reach USD 363 million at a CAGR of 5.2%. Pyrrolidine-3-carboxylic acid derivatives are important molecules as bioactives, catalysts for chemical transformations, and their building blocks. Some of these bioactives have been used in therapeutic pharmaceuticals such as enzyme inhibitors. However, previously known synthesis methods are very time consuming and involve complicated pathways. Here, we present a promising beta-proline synthesis method developed by a group of researchers led by Prof. Fujie Tanaka. The developed technology overcomes the above problems and provides a short and simple 2-step method unlocking new derivatives.

Technology

Essentially, Prof. Tanaka has developed a novel chemically and biologically important pyrrolidine-3-carboxylic acid derivative synthesis method. Highly selective organocatalytic Michael reactions using market accessible starting materials of enoates and nitroalkanes that are useful for the synthesis of pyrrolidine-3-carboxylic acid derivatives have been discovered. Using the Michael reactions, synthesis was achieved in as few as two short and simple steps at low temperature.
