Dynamic regulation of inhibition of T cell activation
Abstract:
Because protective immunity is significantly mediated by activated effector T cells, the regulation of T cell activation is critical for immune response. T cell activation is induced at TCR-microcluster (MC) and is positively and negatively regulated by co-stimulation receptors and intracellular adaptors. Here, we describe dynamic feature of inhibition of T cell activation by recruiting negative regulators in TCR-MC. We first present the dynamic regulation of inhibitory co-stimulation receptor PD-1 mediated suppression of T cell activation through recruitment of phosphatase SHP-2 to TCR-MC. Second, an adaptor molecule CIN85 mediates inhibition of T cell activation by recruiting the phosphatase Sts-2 into TCR-MC. Third, analyzing a phosphatase PTPN22, which is most associated with various autoimmune diseases, inhibition of T cell activation is mediated by recruiting “inhibitory clusters” composed of various inhibitory molecules to TCR MC.
