Cutting-edge Basic and Translational Research in the Carbon-Ion Radiotherapy
Abstract:
Particle beams have predominant advantages over the conventional photon beams for
treating deeply placed solid tumors, particularly effective in killing radioresistant tumors.
Over the last few decades, more than 11000 patients with various cancer types were
treated by carbon-ion radiotherapy using Heavy Ion Medical Accelerator in Chiba
(HIMAC), and we achieved promising results. Here, I’d like to share our recent new basic
and translational findings about the effects of carbon-ion beams alone or in combination
with DNA damaging drugs (gemcitabine or cisplatin) or with micro RNA 200c mimic in
targeting radioresistant cancer stem cells (CSCs) in vitro and in vivo. We estimated that
the relative biological effectiveness (RBE) values for the carbon-ion beams relative to Xrays
at the D10 levels for CSCs derived from several human cancer cells were 2.1-2.4.
The colony and spheroid formation ability of CSCs was markedly diminished by carbonion
beam combined with DNA damaging drugs or with micro RNA 200c mimic. Carbonion
beams effectively destroyed pancreatic or breast or mesothelioma xenograft tumors
when combined with DNA damaging drugs. Taken together, carbon-ion beams combined
with DNA damaging drugs or with microRNA 200 mimic have more power to eradicate
pancreatic, breast and mesothelioma CSCs compared with carbon-ion beams alone.
