Metabolic profiling for cohort and clinical study using mass spectrometry.

Abstract:

Genome, transcriptome, proteome and metabolome are the elements of constituting central dogma, and understanding of the phenotype of an organism. Metabolomics is a promising approach in the search for disease biomarkers because the metabolite concentrations of body fluids are considered as quantitative traits that can describe and define phenotypic characteristics of each individual, which are generated through interactions between genes and environmental influences. I would like to talk about: 1) The establishment of a protocol for metabolomics in a large-scale cohort study with the analysis of human plasma samples obtained from our cohort study “Tohoku Medical Megabank Project” to create a reference “Japanese Multi Omics Reference Panel, jMorp”. 2) A clinical application of high-definition mass spectrometry technology to identify the metabolites in hepatocellular carcinoma (HCC) tissue using UHPLC-QTOF/MS and desorption electrospray ionization (DESI)-MS imaging. We demonstrated that the triglycerides were localized in the tumor and non-tumor regions of HCC by DESI-MSI analysis.
