Description

[Abstract] The actin network generates forces while receiving forces from outer environment. Within the network, actin polymerizes and disassembles continuously. I will present our findings of fast mechanotransduction systems such as actin filament regeneration and stabilization by Formin homology proteins (1-3) and the Brownian ratchet-based force sensor at the tip of thin lamellipodia (4), both of which would not be recognized without real-time monitoring of single-molecule dynamics. I also introduce a new-concept super-resolution microscopy IRIS, which delivers high-definition image with high-density labeling, potentially resolving the problem of inhomogeneous staining (‘Super-resolution Dilemma’), and enables unlimited multiple staining in a single specimen (5).
