Abstract:

Electron microscopy is currently the only available technique with a spatial resolution sufficient to identify fine neuronal processes and synaptic structures in densely packed neuropil. For 3D reconstruction of neuronal synapses, serial transmission electron microscopy and serial block-face scanning electron microscopy have allowed us to examine how experience and neurological disorders change synaptic structures and connectivity at nanoscale resolution. Here I introduce how we applied 3D electron microscopy to synapse remodeling research of motor skill trained rats and genetically modified mouse models. This approach will generate invaluable information on organizational features of neuronal connectomes as well as diverse neurological disorders caused by synaptic impairments.
