Title:
Mitochondrial contribution to dendritic outgrowth in cerebellar Purkinje cells



Abstract:
Cerebellar Purkinje cells develop highly complicated dendrites filled with numerous mitochondria, which provide a large amount of ATP with oxidative phosphorylation in order to maintain the activity of several tens of thousands of synapses. Developing Purkinje cells are thought to require a greater supply of mitochondria for fueling ATP in growing dendrites, which by adulthood can span tens of times as wide of an area as the cell body. However, the detailed molecular mechanisms underlying how mitochondria are linked to dendritic outgrowth are not fully understood. It is known that mitochondrial fission is important for the proper distribution of mitochondria in dendrites, and that mitochondrial biogenesis is the process by which cells increase their individual mitochondrial mass and activity to increase ATP production. We investigated how mitochondrial fission and biogenesis contribute to a stable supply of ATP for the greater energy expenditure in developing dendrites.

In this talk, I will present our research focusing on the function of mitochondrial fission protein Drp1 (dynamin related protein 1) and mitochondrial biogenesis regulator PGC-1α (peroxisome proliferator-activated receptor gamma co-activator 1-alpha) during dendritic outgrowth in cerebellar Purkinje cells.
