Abstract:
Operons are a fundamental feature of prokaryotic gene regulation. However, how they form through evolution is yet to be understood. Through an attempt to mimic the genome evolution of bacteria living in insects, we came up with a new model of operon formation: mobile genetic elements called insertion sequences (IS) brings together two genes placed apart to form operons. We constructed a fluorescent reporter system, introduced it into Escherichia coli, and evolved it in the laboratory, showing that new operons can form by the proposed mechanism. Our study implies that genomes of modern prokaryotes have been shaped through evolution under coexistence with mobile genetic elements.
