Title: The dentate-hippocampal network support spatial working memory.
Abstract: The dentate-hippocampal network plays a crucial role in working memory. To understand neurophysiological mechanisms underlying memory function, we lesioned dentate granule neurons and monitored the activity patterns of hippocampal CA3 circuits as animals solve a dentate-dependent spatial working memory task. We revealed that ripple events in CA3 were specifically increased during reward period, which are predictive of behavioral performance. The reward-associated ripples were abolished specifically at recording sites determined to lack TIMM-positive mossy fiber projections. The reduction in reward-associated ripple events was accompanied by a selective decrease in slow gamma power. CA3 cells with no mossy fiber projections showed decreased spatial tuning and less proportion of prospective firing patterns during reward. The direct input from dentate granule cells to CA3 may be critical for spatial working memory through the dynamic regulation of a distinct class of network activity.
