Title: Expanding the Nucleic Acid Chemist's Toolbox: New Fluorescent Cytidine Analogues

Abstract: Nucleoside analogues that possess fluorescence properties which are sensitive to their immediate environment have found many uses in nucleic acid chemistry. We have had a long standing interest in structurally compact, base-pairing competent, and hybridization responsive nucleoside analogues that are intrinsically fluorescent, that is they do not rely on an appended fluorophore. Nucleoside analogues with such properties have a variety of uses including the ability to act as reporter groups for nucleic acid-enzyme interactions and as sequence probes.

Much of our work has focused on cytidine analogues particularly pyrrolocytosine (pC) which is a convenient scaffold for luminophore development because substitution of the pyrrole ring strongly influences the fluorescence. An overview of our work with pC and demonstration of some selected uses in DNA, RNA and PNA will be presented along with our recent developments.
