Title: Transgenerational Inheritance of Stress-induced Epigenome Change
Abstract: Transgenerational effect of stress has been longstanding interest, because it affects phenomena ranging from disease to evolution. For instance, it has been suggested that nutritional stress during early life increases the frequency of life-style-related diseases, such as diabetes, which could be inherited (Barker’s hypothesis and DOHaD hypothesis). Some observations suggest that the stress-induced epigenetic change is inherited, which partly resembles to the inheritance of acquired characteristics. However, insight into the underlying mechanisms is very limited.
Recently, we demonstrated that ATF-2 family transcription factors mediate the stress-induced disruption of tight heterochromatin structure, which can be inherited to next generation (1, 2). ATF-2 is a member of the ATF/CREB superfamily, which was originally identified in our lab (3). ATF-2 is phosphorylated by p38, a stress-activated protein kinase, in response to various stresses, including environmental, nutritional, psychological, and pathogen infection stress. Our recent data suggests that those various stresses induce epigenome change of ATF-2 target genes, some of which can be inherited to next generation. I will discuss the relationship of this phenomenon and some diseases.
