Title: Digital counting of mRNA molecules genome-wide by molecular barcoding

Abstract: Determining the absolute copy number of molecules (e.g. copy per cell) system-wide accurately is important for understanding what is going on in a cell at the molecular level, for integrative interpretation based on multiple omics alanyses, e.g. proteomics or metabolomics, or for quantitative mathematical modeling which relies on quality of raw data.  We have been developing a method which enables the digital counting of the absolute copy number of mRNA molecules genome-wide from limited number of cells (≦100)  by using originally developed molecular barcoding.  This highly accurate quantification allows us to distinguish a small difference of mRNA expression genome-wide including low copy RNAs, which are important since the expression level of transcription factors that play crucial roles are thought to be low.  We applied this method for limited number of mammalian primary cells in order to study, for example,  differentiation in hematopoietic system.  I will present our technical development which also provides higher dynamic range for the quantification and its applications including single cell analysis.
